Advice on high-end light-sheet

Hi,

We are considering several options for a high-end light-sheet (lattice)
system and would like to know your experience. The system is meant for
our advanced light microscopy facility at the CBMSO in Madrid, with many
users and projects from different fields of the biosciences. So we value
versatility and robustness, in addition to specifications.

I would be very grateful to listen to your comments off-list (if you
think it should be that way).

Versatility?
A good microscope should be able to deal with any sample you throw at it,
right?
Not so with a lightsheet microscope. You need to decide whether you need
large sample volumes (Lavision Ultramicroscope, Zeiss Lightsheet Z.1 - now
apparently rebranded as " Lightsheet 7") or high resolution (3i lattice
lightsheet or Zeiss "Lattice Lightsheet 7" - how confusing).
If you want both options, you're going to need two separate instruments...
Absolutely zero commercial interest.

Hi, well, a great answer from Zdenek.
The Zeiss Lightsheet 7 can do both large samples (think cleared mouse brain) and small samples (think small transparent organoid), but you need two completely different stage set-ups.
Bruker simply advises you to purchase two separate systems. Maybe a good idea.

Versatility is not the strong point of a lattice light sheet.
Would I like to have one? Sure, however I think it would be difficult to justify based on the number of samples that could be imaged.

Very clear and sound!

For a better understanding from my side and context, please translate
"versatility" as "accommodation to as many different applications and
user requests as possible". And yes, you are right on the size of the
sample.

As I understand it, most of our users would be more on the gaussian side
of light sheet (bigger samples, embryos, etc), and I was wondering what
would happen with those interested in imaging medium-sized samples, such
as organoids, etc. Lattice light sheet will surely be very kind to
cultured cells and much more resolutive. But for now, we have other
instruments (spinning disk, STED) that cover a wide range of
applications in this category.

As in my earlier email, I'll be very interested and appreciate any
experienced input.

We have an Aurora light sheet from MSquared Lasers and are delighted with it. It is very versatile and with its Airy beam, offers high res across a large (around 800 um) FOV. :)
