Dear All,

We are considering to purchase (write a proposal) for a light sheet system. For the completeness of the provider list I wanted to ask you whether you  are aware of any other commercial system then the one from: Zeiss Z1), Leica (DLS), Luxendo, I3 (based on the ASI setup) and LaVision (Ultramicroscope).
I know that there are considerable differences between the application fields (sample mounting etc.) of these different systems, but wanted to make sure that I have the complete list. Furthermore, is there anyone who have compared the Leica solution to other ones? I would very much appreciate if you could share your feedback either on the list or privately.

Thanks Gabor

Hello Gabor ,

The ALPHA3 system from Phaseview ( France) is another option for light sheet microscopy recently launched on the market but with some clear distinctions from other existing systems.

You should check the following link : http://www.alphalightsheet.com

We are the distributor in the US and Canada, for other territories, please contact Phaseview SA directly !

Hi Gabor,

Here’s my list of commercial light sheet systems that I’m aware of. There are a few that you didn’t mention.

- Zeiss Lightsheet Z1
- Leica SP8 DLS
- ASI diSPIM
- ASI oSPIM
- 3I Marianas
- 3I Lattice Lightsheet
- LaVision Ultramicroscope
- Cairn L-SPI Illuminator
- Luxendo MuVi-SPIM
- Luxendo InVi-SPIM
- Phaseview Alpha3

Those are the future setups that I know of (in case you have time to wait):

- Zeiss Lattice
- Leica SCAPE

I got a first impression of the Leica DLS last year, and it’s definitely interesting. You need a full confocal to get the light sheet illumination, which might be a pro or con. Personally, I’d like to have both techniques in separate setups, as they serve different purposes. It would be nice to have a simplified, matched scanhead without detectors, just for light sheet illumination. One could still use it for sequential photo-manipulation and light sheet imaging.

Best,
Michael

Hi Gabor,

Cairn Research (UK) produce the L-SPI (L-shaped single plane illuminator).
It's completely platform-independent and can be adapted to diverse
experimental settings. It's based on a prototype I made for looking at
corals, so it works well with large, photosensitive samples. Papers on the
setup and applications should be published soon. Until then, please feel
free to contact me offline for more information.

With kind regards

Philippe Laissue

Note: I work with Cairn Research in the context of my industrial
fellowship, but I have no financial interest in the L-SPI. I'm just jazzed
to see it get used:)

A very good list!

Just to add to it, I believe that M2 Lasers (M-Squared Lasers) also has an Airy light sheet. They don't seem to have it on their website, but they were publicizing it at BiOS.

We've used the DLS and the Z1 to look at agar enclosed spores and cleared lung samples. The DLS needs to have your sample mounted on a small dish, whereas the Z1 needs you to suspend your sample into their enclosed chamber. They have capillary tubes, butcher hooks. With the Z1 there is scope to 3D print your own stage holders. Something to think about.

With the DLS you have the potential to combine LSFM with traditional confocal microscopy, so for example you could bleach/ablate a region with the confocal set up, then switch to LSFM to monitor the whole system.
I think the DLS comes in at ~£70 000 as an add on to your confocal, whereas the Z1 may be ~£300 000 I think...(please correct if I'm wrong)

They are the only systems I've had experience with.

Please account for significant investment in the PC and storage side of things. These things spew out data at an alarming rate if you're not prepared.

Happy hunting!

Darren

Dear All,

Thanks for all of you who shared information about the light sheet systems! I learned again a lot, this list is an amaring resource.
