FV10i opinions? (for a facility)

Greetings to all

The microscope facility is evaluating the purchase of an Olympus FV10i (confocal
in a box). This equipment would be used for "standard" users (488, 543 and 633
nm) in order to reduce the work load on our Zeiss 510 Meta.
I would appreciate all your opinions, and if possible, the price range to which
you have been subjected (either at purchasing the equipment or as users at a
facility). Please feel free to reply inter On or Off-list.

Dear Alejandro,

the Olympus FV10i is a very nice instrument for routine work. Thus if you know that you are doing the same experiment the next years this is exactly the machine you want to buy. My experience in a facility is that you (your users) sometimes need more flexibility. And this is only partially offered by this microscope (in terms of optics but also/especially in terms of software). Therefore I would not consider this machine for a facility unless I know that I have a number of users which only need the capabilities of exactly that machine now and presumably also in the future. I'd prefer to buy a Leica SPE, Nikon C1 or Zeiss LSM700 for a facility.

Just my 2c.

Hi Alejandro,
   In line with the previous reply.  We just purchased a Zeiss LSM700 for
our lab and I have to say this is a really nice instrument and will be great
for "standard" users in a core.  I know price is always a consideration, but
you might be surprised at what Zeiss offers this system for, so it is worth
a look.

Dear Alejandro,

We have also tested the water-based FV10i-system and I totally agree with
the other repliers. The major drawback of the setup is lack of optical
flexibility (the optical zoom option cannot replace individual objective
solutions). The system can be highly recommended, however, for confocal
beginners.
What I liked is the stability of the closed-box system for long-term
experiments and its focus stability during multi-positioning. Moreover, the
self-dispensing option raised no problems with water evaporation during 2
1/2 days of imaging, though at low-speed (definitely the system cannot be
used for fast acquisition).
What I missed was the option for plate screening and that the oil-based
system also includes environmental control.
In case the facility users do a lot of easy routine jobs you could buy one,
although I am in line with Arne that for some 30-50k€ more you will get a
more flexible system from Zeiss or Leica.

Does anyone have any experience upgrading a 543 HeNe to 561 DPSS laser
on a Leica SP2 AOBS confocal? I have inquired to Leica Canada and
received the following. "In regards to the 561 laser for your
microscope, we estimate that the laser will cost about $40K plus labor
for the upgrade."  This cost seems very high to me.

Hi Mike,

We are doing a similar upgrade... One additional 561 DPSS on SP5.. Initial quote was 76k USD  :(

In the end we bargained a bit, but still very expensive. Everything has to be shipped from Germany. Installation takes 4 working days. :((

Dear Michael,

The price for the 561nm upgrade seems to be correct - we paid approx.
24000 Euro two years ago (1 Euro ~ 1.3 Can.$), not including labor. But
the advantage is a laser that excites commonly used red fluorophores
with higher efficiency. Plus it has more power and does hopefully not
age as fast as gas lasers.

We upgraded 543 HeNe to 561 DPSS laser on a Leica
SP2 AOBS confocal three years ago. It cost 30.000
€ . It works much better than  543 HeNe. It´s worth.

Hi Micheal,

we just ordered the 561 DPSS upgrade. It took quite some bargain to get
around 24K (VAT not included). We are now waiting for the laser to be
installed.
