Glass bottom 96 well plates for hi-res imaging

Hi all,

We have a user who is after some 96well plates for hi-res imaging, ie coverslip thickness (glass?).

Any comments/recommendations would be appreciated.

Hi Adrian,

I've had great success with plates from Matrical.  They sell #1.5 96-well plates with square wells, which maximises the number of cells you can image per well.  We found that they were more consistent than the Whatman plates and my cells grew much more happily in these plates.

We have 'recently' (several months) encountered great problems with the Matrical glass bottom 96-well plates. After using them for years without any trouble, the cells suddently started dying (all sorts of cells, any type of medium, in 2D or grown in a gel) as soon as they were seeded. Matrical claimed that they had not changed anything then they blamed the glue but the next batch of plates has the same problem.

We had a similar problem with Povair plates.  The problem could be resolved with a methanol wash, though admittedly this isn't very practical with a large number of plates.

Our customers had good success with Greiner Bio-One coverglass bottom plates, although I was not able to find them on their online catalog in a recent search.

I've had good recommendations for Ibidi plates and Willco wells plates. They are supposed to be good optically, and cells are reported to grow well on them:

Although not coverglass bottom, optical plates from BD and Nunc have also given us good results. They do work well with lenses such as 20x/0.8 PlanApo, but we did not use them with higher NA immersion lenses; BD used to have also glass bottom plates (#357311); I'm not sure they still do. The BD Thin Bottom series (#353219) are "optical grade" plastic 190 microns thick. BD claims they are suitable for use with high NA objectives. Assuming the refractive index is right, oil immersion lenses shoudl not care too much about a 20 micron difference in thickness.

I've never used them (nor do I have commercial interest) but you can
get a free sample from ibidi: http://ibidi.com/freesamples.  The
bottom is "coverslip-like #1.5" plastic.  I have used their chamber
slides with no problem.

Hi Adrian,

One thing you may want to check is the position of the bottom of the wells
compare to the plastic frame. The greigner's one I had first were not
flush  with the frame and thus I had issues with the objective touching
the frame when imaging the wells on the edge. So I always had to sacrifice
the edge wells and decided to switch to the Matrical plates.
