What is the .lifext file for and do I need to keep it? Has anyone got the stage overview option and do they find it useful? In particular, does it all you to set up a non rectangular tilescan (ie just pick the area that you actually need / want to image?) When you zoom in, the system now interpolates / antialiases / smoothes by default. I can turn this off for an image but can it be globally disabled? Has anyone experienced system lockups where the laser excitation just stops part way through acquisition or live view and the system thinks that it is still acquiring and locks up forcing us to do a force quit via task manager? Thanks in advance, Dave Johnston, BIU Southampton. |

You can load default sample carriers, register them, take overview images (including a very useful spiral view) and then draw arbitrary shapes on the preview to define your tiles, tile sizes adjust when you change objectives, you can add positions and so on, click at any point to get a live view. I guess with a confocal it would be good to have a resonance scanner to work efficiently. We have similar functionality with the Zen blue software from Zeiss for the widefields, but not for the Zen black confocal package. best wishes Andreas -----Original Message----- From: Dave Johnston <[hidden email]> To: CONFOCALMICROSCOPY <[hidden email]> Sent: Sat, 2 Sep 2017 8:14 Subject: Leica SP8 LAS-X features ***** To join, leave or search the confocal microscopy listserv, go to: http://lists.umn.edu/cgi-bin/wa?A0=confocalmicroscopy Post images on http://www.imgur.com and include the link in your posting. ***** Hi, we have just had our SP8 PC system rebuilt to try to cure crashing issues and the rebuild has included the latest version of LAS-X which seems to have extra features which I can't find documentation for (not in the release notes for the version or help files or online) and I would be grateful for comment. I think if you go to file>help it opens a nice index of every feature. However, we also had to spend a lot of time figuring out the files system upon the update. Wait until you discover the .lof autosave formats, which for LASX is ok, but unreadable in FIJI/IMARIS. Best stick with TIFF there. One thing to watch with the stage overview is if you click far away from your positions the stage will zoom there. Very scary if your finger slips on the mouse and the objective is in-focus! Our SP8x crashes sometimes if we save a data set while acquiring, so maybe the locking is occurring when you're trying to do something else?
