Dear Listers,

One of my colleague is interested in staining only the lipid containing
vesicles in the C. elegans. She want to stain these vesicles specifically.

Are there any dyes / tags which can be used for this purpose.

Any help on this issue is welcome....

I think there was some work using Nile Red for this. I'm not sure how
specific it is though, you'll have to check the literature.

We often used BODIPY (for microalgae). It stains the lipid bodies very well.

Dear Deepak,

if your colleague tries to stain lipid droplets -not vesicles in the strict
sense- these are specifically stained with BODIPY or NileRedO. CARS is
another very good option if you have the tools.

Hi Deepak,
I found this article by Eyleen et al. 2009 (C. elegans Major Fats Are Stored in Vesicles Distinct from Lysosome-Related Organelles, Cell Metab. Nov 2009; 10(5): 430–435). The article describes methods of using NileRed and BODIPY for staining fatty acids. Hope this helps and good luck.

Third Harmonic Generation (THG) would be another multi-photon technique
to do image fat droplets. We had nice images if the worm was not moving
(i.e. fixed) when we tried that for a practical course
