Multiphoton spectra of fluorescent dyes

Greetings,

For one photon excitation, the excitation and emission spectra of commonly
used fluorescent dyes can be viewed by using the following tools:

* Invitrogen Fluorescence SpectraViewer, www.invitrogen.com

* BD Fluorescence Spectrum Viewer, www.bdbiosciences.com

I am wondering if we have similar tools for fluorophores with multiphoton
excitation.

Hello Gary,

I have found the DRBIO website (Developmental Resource for Biophysical
Imaging Opto-Electronics at Cornell- Warren Zipfel, Watt Webb and crew
) at http://www.drbio.cornell.edu/cross_sections.html to be an
excellent resource for at least the 2P excitation cross sections. The
following paper is good commonly used reagents. I have yet to find
anything equivalent to the resources you mention however.

relevant for cell imaging.

Bestvater F, Spiess E, Stobrawa G, Hacker M, Feurer T, Porwol T,

Good afternoon,

while I am not aware of a similar on line tool for multi photon
excitations, there are publications, which list "MP" excitation spectra of
a number of dyes, e.g. by Bestvater et al.

Gary,

Have you tried the DRBIO resource at Cornell that is run by Warren Zipfel now? They have a list of two-photon absorption cross-sections there that I find helpful. Its not exhaustive, but they have a number of the fluorescent proteins and alexa dyes listed there for example, and you can download the ascii data as well.

Probably the most comprehensive site would be at: http://www.spectra.arizona.edu/ .  We've compiled all the 2P data we can find, including the DRBIO data.  Simply select "2-photon cross-sections" under Dye category.  We hope to incorporate the ability to download any data the user chooses, soon.  This site has been assembled by Dr. Urs Utzinger at the U of A Biomedical Engineering Dept. and myself, with the code-writing help of Gary Carstensen in our Instruction/Access office, and includes much of the 1P data collected by Dr. George McNamara, U. of Miami, and myself.

Additional spectral data from optics, dyes, filters and light sources always welcome.
