When we grow them in petri plates it spreads very well but when seeded on coverslips it remains rounded. What could be the possible reason. Cells usually don't like hydrophobic ("dirty") glass slides. A short (oxygen) plasma treatment could help. Of course there are also many solvent based cleaning protocols around. The normal lines didn't seem to care whether they were on glass or plastic. At least they looked the same - we use a coated coverslip now. I vacuum deposit a metal (platinum is good, germanium, or silicon itself seem to work about as well). Then, the cells treat the surface just like tissue culture plastic. Get the RS, which has a coating that resembles tissue culture plastic. Usually, coating the cover slips with a substrate like poly-D-lysine or collagen will take care of the problem, but there are some cell lines that will never grow as well on glass as plastic.
