PFS or similar on upright

Dear all.

 

We are looking for 2-photon application equipment with an upright microscope. Is it possible that this upright microscope has a PFS or similar system?

Hi Konstantin

 

The hardware autofocus will only work if you have a coverslip and your sample is a fixed distance from the coverslip. This is designed for inverted microscopes. I do not think it is possible to use a hardware autofocus system on an upright microscope.

Hi Konstantin,

One way of doing that would be to combine an inverted base that has hardware autofocus with an upright upper part that is your actual microscope. Then you could use the hardware autofocus to keep a dish at a fixed z-position. I’m not aware of a commercial implementation, though. Also keep in mind that commercial autofocus systems (at least the Nikon PFS) use 840nm, with which your 2P excitation might interfere.

Try ASI CRISP modules that goes into the C Mount of upright scopes. Works well with coverslips n they have micromanager plug in which you can work independently of your 2P imaging software.
