Dear list,
Does anyone know what type of plastic can be used for 3D printers to print sample holders that will be used for iDISCO sample imaging in organic media, such as Eci or DBE?

Thanks a lot for your help.

Hi Verona

Silicon works but it is soft so you need a plastic holder.

Hi Verona,

There are several suggestions (some mine, some from other folks) here: https://forum.microlist.org/t/any-experience-with-solvent-resistant-materials-for-sample-holding-in-lightsheet/89

Happy to discuss more or provide designs/advice off-list.

Best,

Hi Verona,

We just used Mattek-style glass bottom dishes.  To prevent DBE from eating the plastic and glue, we cut the neck of a volumetric flask into short cylinders with the diamond powder cutting tip on a Dremel tool.  Then we used the same tool to smooth the bottom of the cylinders and secured the cylinder to the dish with superglue.  Any cyanoacrylate glue is compatible with DBE.  If you have a 3D printer and the appropriate plastic type then that may be easier; this was just the fastest route for us.  All the best,

T

I haven't had any problems with ECi melting through any plastics to date or
harming any dipping objectives (have not tested it on polystyrene tissue
culture plates). We have converted most of our iDISCO users to ECi rather
than DBE and have not seen any decreases in performance. However, it is a
very limited range of fluorophores that have been tested to date.

BONUS: the facility now has a very pleasant cinnamon smell most days.

Yuck! I don't like that sticky smell! ☹
Yes ECi dissolves polystyrene pretty quick but not polypropylene.

A strategy is to print with cheap plastic, then dip in or paint with a
silicone resin to coat and protect it.
