To prevent DBE from eating the plastic and glue, we cut the neck of a volumetric flask into short cylinders with the diamond powder cutting tip on a Dremel tool. Then we used the same tool to smooth the bottom of the cylinders and secured the cylinder to the dish with superglue. Any cyanoacrylate glue is compatible with DBE. If you have a 3D printer and the appropriate plastic type then that may be easier; this was just the fastest route for us. We have converted most of our iDISCO users to ECi rather than DBE and have not seen any decreases in performance. However, it is a very limited range of fluorophores that have been tested to date. BONUS: the facility now has a very pleasant cinnamon smell most days. -Doug On Mon, Nov 25, 2019 at 11:44 AM Ariel, Pablo <[hidden email]> wrote: > ***** > To join, leave or search the confocal microscopy listserv, go to: > http://lists.umn.edu/cgi-bin/wa?A0=confocalmicroscopy > Post images on http://www.imgur.com and include the link in your posting. > ***** > > Hi Verona, > > There are several suggestions (some mine, some from other folks) here: > https://forum.microlist.org/t/any-experience-with-solvent-resistant-materials-for-sample-holding-in-lightsheet/89 > > Happy to discuss more or provide designs/advice off-list. > > Best, > Pablo > > Pablo Ariel, Ph.D. > Assistant Professor > Director of the Microscopy Services Laboratory > Brinkhous-Bullitt Bldg B04 > Department of Pathology and Laboratory Medicine > University of North Carolina at Chapel Hill > http://www.med.unc.edu/microscopy > Tel: 919-966-2413 > > > -----Original Message----- > From: Confocal Microscopy List <[hidden email]> On > Behalf Of Villar Cerviño, Verona > Sent: Monday, November 25, 2019 9:36 AM > To: [hidden email] > Subject: Plastic for sample holders for iDISCO+ sample imaging > > ***** > To join, leave or search the confocal microscopy listserv, go to: > http://lists.umn.edu/cgi-bin/wa?A0=confocalmicroscopy > Post images on http://www.imgur.com and include the link in your posting. > ***** > > Dear list, > Does anyone know what type of plastic can be used for 3D printers to print > sample holders that will be used for iDISCO sample imaging in organic > media, such as Eci or DBE? > > Thanks a lot for your help. > Best regards, > > Verona Villar, PhD > Servicio de Imagen > Instituto de Neurociencias CSIC-UMH > E-mail: [hidden email]<mailto:[hidden email]> > Tlf: +34 965 91 9266 > http://in.umh-csic.es/es/si_presentacion.aspx > |
