Slidebook

Search the CONFOCAL archive at http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal Does  anyone successfully and happily use Olympus' Slidebook?  We have had a devil of a time with automatic image normalization (making controls look as bright as positives), merging images and just image file handling.  Any expert help would be greatly appreciated.  We are just hating this program.

Hi Elaine,
The normalization occurs in the display, and does not affect the  
intensities saved to the native Slidebook file.    Most acquisition  
software does this, some  automatically set the exposures so that the  
images themselves are falsely bright.  I've not found  a preferences  
setting in Slidebook for a non-stretched default view.  However, if  
you open the normalization window, the lower  button  will normalize  
all channels to the lowest and brightest intensities, preserving the  
relative intensities in an exported RGB TIFF, although this does not  
set all channels to 0 and 4095.   If you were wanting to export the  
Slide files for deconvolution or densitometric analysis in some other  
application, you would export in 16-bit which uses native intensities  
without any histogram stretch.
