Hi!

Does anyone have any protocols for fixing and staining actin in whole plant roots that works really well?

All suggestions are appreciated!

Thanks!
Terri

Check out David Collings' papers. I think this one has intact root double labelling of actin and tubulin:

Collings, D.A. and Wasteneys, G.O. (2005) Actin microfilament and microtubule distribution patterns in the expanding root of Arabidopsis thaliana. Canadian Journal of Botany 83: 579-590

Terri,
In addition to the paper that Rosemary White mentioned, you
can also check:

Rahman et al. 2007 Plant Journal 50: 514-528.   THis protocol is a
bit complicated but I do feel that it works well. If you have
specific q's feel free to email me off line.

Good luck,
        Tobias
