UV laser advise for Zeiss 510

UV laser advise for Zeiss 510

Dear List,

The original 405nm laser (possible iFlex...from PointSource Ltd) in our Zeiss LSM510 was dead and replacement cost (quotation from Zeiss) was out of my imagination.

Does anyone has any suggestion on 1) where to get a similar/alternate part (but need to physically fit to the space), 2) setting up a totally different UV laser (such as Enterprise, 360nm etc, but need to be compatible with AIM software 3.2?), or 3) surplus/on sale LSM510 that the parts can be recycled?

I have noticed a similar posting two years ago by Josef, but speculated there might be some changes in term of product availability and/or technique advance.

The technology for the 405nm lasers has progressed significantly and the
prices have come down drastically.  Unfortunately the microscope
manufacturers don't seem to have caught up with this trend yet.  There are
three points to consider when choosing a replacement laser: 1) Power
Stability  2) Mode quality  3) Power output/lifetime
There are many third party vendors that can meet your specs for various
prices, so do your homework.

Hi Guosheng,

My former LSM510's Coherent Enterprise I UV (351/364 nm) laser is
probably still on the shelf at Laser Innovations
http://www.laserinnovations.com/home_LaserInnovations.htm
They can refurbish and ship to you everything (laser head, power supply
and water chiller). I still have the UV "coupling" component - can
arrange purchase if that is the way you want to go. We were using the
old LSM software (3.2 sounds familiar).
Keep in mind that it might need another round of refurbishing in after
less than 2 years of use.

For now, you should be able to acquire DAPI on your LSM510 by using the
widefield DAPI filter cube with the pinhole wide open. If you have a 633
nm laser, you can use the "Cy5" channel with To-Pro-5 or other NIRF dye
- could buy a lot of to-Pro-5 for the cost of buying a UV laser. Users
could still counterstain with DAPI for looking by eye, but add in
to-Pro-5 for confocal specimens.

Within the year(s) nanoscopes may go very low price - practically the
cost of ~1 Watt lasers and gizmo like an HDTV format DMD (I was told
Texas Instruments has a black background HDTV DMD development kit for

Shroff Nature Methods paper on MSIM. Alternatively, STORM may take over,
as in PubMed  22891300.
