What did you do with your old LSM510?

Hi all,

Ours ceased working because of yet another card breakdown and we do not
feel it's worth putting another 5k EUR into it. Again. So we decided it's
time.

Did anyone try to convert it to a wide-field fl?

Hi Avi,

Yes, we converted the Zeiss LSM510 Axiovert 200M inverted microscope I
managed at UMiami to widefield (internal motorized Z, manual stage).
Went fine. This was several years ago, so a CCD camera was used.

We are in the process of converting our 510 to a WF scope with motorized z and motorized stage using Metamorph to drive it. We previously did this with our 5 Live scope (We now have two SP8's - one with MP, FLIM/FRET and 3 color STED, the other with digital light sheet plus a separate GSD system. Life is good!).

Anyone interested in the lasers/laser control box scan head for parts can contact me.

Hi Avi,

Indeed, we also converted one very old 510 into a pretty nice wide-field
scope. It is one based on the Axiovert 100M and I found that
Micro-Manager (https://micro-manager.org/) drives all the components on
that model scope, including the dichroic block mover. Add a decent
camera and good to go. (btw. Axiovision also drives all components on
our scope but M-M gives more flexibility in camera choice).

Hi Damir,
Did you get  help from reps, or did you manage to do this on your own. What
about connection to the MM computer?
 I have not opned it up yet, but everything seems go through the giant
laser box thing. Is it a matter of just connecting cables to a computer?

Hi Avi,

The real time computer (has a bunch of aluminum slats on the front) is
for the confocal components only. Just disconnect from the Axiovert the
scan head and any cables that go to the scanhead and real time computer.

If there is another Aciovert 100 on campus you might compare cabling
with that, and maybe even try Micro-Manager on that first.

Hi Avi,

As George says, the huge box with lasers, power supplies, RTC, etc. all
become surplus and any cables between the Axiovert and that box can be
disconnected. I look briefly on your facility's website and noticed that
your LSM 510 is built on a Axiovert 200M. That's a great scope and very
well supported by MM through the ZeissCAN adapter (not the ZeissCAN29
one!!). All your motorized components will be controlled by MM. A few
things I note from looking at the picture on:
http://lifefaculty.biu.ac.il/FacultyInstruments/index.php?option=com_content&view=article&id=2&Itemid=71
- I cannot see where the META scanhead is connected. Is the right port?
I don't know how to properly defeat the laser interlock on a 200M so you
may need to ask the Zeiss technician for help on that.
- It appears that you don't have a scanning stage but if you do, its
controller (MCU27 or 28) will need to be plugged through a CAN bus cable
into the back of the Axiovert 200M. So, a single RS232 serial cable is
all you'll need between the Axiovert and the MM computer.
- It looks like you have a transmitted PMT. You'll need to take that and
the switcher off of the transmitted light pillar and mount the
transmitted light lamp directly onto the back of the pillar.

Hi Avi,

we just did this and as the other replies say, its is easily possible to use the µManager or AxioVision software to control the microscope via the serial port.
HOWEVER, in our case (LSM510 META with Axiovert200M  from 2006) we had to remove an additional safety shutter behind the eyepieces. This shutter could NOT be controlled by any other software but can be easily blocked in open position or completely removed. I did it myself, no need to call the service. If you need more infos/instruction how do do it, you can contact me off list.
