Zeiss LSM710

I was wondering if people have one or have done extensive demos on the new Zeiss LSM710. I would be interested in general feedback about the system and its performance. I am particularly interested in sensitivity and the new spectral detectors.

Leica DMI6000 perfusion system

Dear all,

 

I write you because I am trying to install a perfusion system in an inverted Leica DMI6000 microscope and I was wondering if any of you had the same problems before.

 

Our microscope is installed in an incubator ML and it is equipped also with a heating insert P (heating stage).

This stage is a solid rectangular piece made in aluminium and is designed with a couple of openings (one in front the other) for the tubes that will allow the medium exchange.

BUT, the system does not come with a pump or the tubes for the perfusion.

 

I contacted Leica to check whether I was missing any parts of the system, but they suggested me I have to install them.

I checked then if the perfusion system FCS2 was suitable for DMI6000 microscopes, and the FCS chamber is for a circular stage, so I can not adapt that to my stage….

 

I was wondering if the solution is to buy a pump and the tubes independently.

 

Does anyone have any idea on how to solve this?

Hi Paloma, 

What are your samples? are they just in a Petri dish  inside your heating insert P? Can you heat the whole incubator, or just the insert?

I can only give you some general pointers. Bioptechs has pumps and other accessories for perfusion. You may be able to build your own system from their components (pump, tubing, and needles), maybe with a bit of help from your mechanical shop:


If that's too difficult, you could get one of their open dish systems, such as Delta T, where there are already holes for the accessories and such. If your incubator is heatable, all you need are the dishes and stage insert. They should have them for most microscope stage fittings.  If you can't heat the incubator, then you would need also the heating control unit... obviously, this would add a few thousand Euros. I have no commercial interest with Bioptechs... just familiar with their products. Other vendors of microscope accessories should have similar products. I am surprised Leica wouldn't be able to help you with that. One European supplier of accessories for Leica and Zeiss microscopes is Pe-Con:



They do have some perfusion accessories which should fit Zeiss or Leica stage inserts (your stage insert P is probably from them).

Zeiss LSM700

I would like to hear from users of the Zeiss LSM700 system.
We are considering the purchase of this system, and I am wondering how users of
this system like it. Particularly, can the continuous emission beamsplitter be
used without  additional emission filters (which Zeiss suggests as a high
sensitivity mode).

We've purchased one and have been using it for a few weeks. It is a significant upgrade from the 510.  More sensitive and the spectral imaging is wonderful.

Have people with the LSM710 checked how close the laser lines the system  can
image? If so, I would be very interested in the results.

I guess that depends on your sample and laser power, during our demo they
imaged right over the laser lines and you could see nothing of the laser,
but this was with one of the usual super bright test samples. In this case
the laser lines are completely filtered out by the dichroics.
I later did some test on individual, multiple fluorophore labelled Fab's
(cy3) attached directly to a coverslip, you loose about 3 images (30 nm), i
had to open the pinhole a bit to get good S/N this might have affected the
laser line suppression. However the signal was impressive for a confocal.
According to Zeiss you can get the LSM 710 with special dichroics for single
molecule detection (FCS) which have double the suppression than the ususal
ones, but this is more expensive.

Optic Fibre Cable - sources?

We have an old MRC 1024 and the optic fiber is shot. Does anyone have a
source for replacing one?

Oz is good for custom stuff.  If you can get away with an off-the-shelf patch it will be cheaper.  Edmund and Thor (among others) sell these.  You just need to know the fiber type and termination options.
