Hi all,

I got a problem with my confocal microscope.
I see in the image some concentric circles.
Any advices to work this problem out?

Newton rings from imaging too close to the coverslip (reflections)?

Dear Nicoletta,
Does this occur with all lenses or predominately low magnification lenses, all wavelengths or more noticeable with short wavelengths?  As Doug said, could be coverslip.  Other possibilities are a DIC analyzer in the light path, double coverslips (it happens), choice of mounting medium or an incorrect or deteriorating dichroic or filter allowing  laser reflections to leak into the detector.

This is interesting. I agree that it might be Newton's rings.   On our
Leica, SP!, which has spectral filters, we often see the rings if we set
the wavelength detector too close to the exciting wavelength.

I have a Nikon C1si Spectral Confocal system on a Nikon TE2000E inverted
motorised microscope, I used a 60x lens and the 488 filter, placed on a Melles
Griot Ar laser 457-514 nm. It didn't happen with other channels but it happened
also with the 40x lens.
I saw the rings just on the upper left side of the image. The weird thing is that it
happened for two consecutive days, and this morning it didn't happen anymore.
That is the setting I usually use. I can say that it happened after I and a collegue
of mine alligned the laser.
