Hello everyone,

We have a Leica SP5 X confocal microscope. We are planning to image  
96-well plates on it. But so far I haven't found anything to fit the  
plate on the stage.

I'm wondering anyone on the list has experiences with this kind of  
experiment. Any suggestion is welcome.

Dear Yue,
Have you contacted the stage manufacturer?   Most motorized stages can accept plates but you may need to buy an adapter from the manufacturer.  Be aware that the distance between the bottom of the plate and the plate's plastic skirt is a few millimeters and varies between plate brands, and styles within a brand. The skirt requires that you bring the object lens so far up to reach the bottom of the plate that the stage can strike either the imaging lens or an adjacent lens, especially when someone else gets on the system with a slide or dish.  If the stage maker does not offer a plate holder that will lower the plate's position to reduce the problem, but you can usually buy a solid blank insert which can be taken to your local machine shop.

No personal experience with it, but Leica and EMBL have jointly
developed a hardware & software package to do that. I've seen a webinar
on it, early last year. Maybe that is still archived somewhere. I
suggest asking Leica...

There is a special stage insert for 96 well plates, I think you need  
to purchase separately if you don't have already.

You need to check the model of your stage then only it will be
possible to search for the 96-well plate holder insert.

Hello,

- Use plates specifically designed for high-content imaging. Typically these have a low skirt,
to avoid a conflict with the objective at the sides of the plate, and a decent optical quality
bottom of the correct thickness.

- On our Leica SP2, we can insert a plate holder in the (Marzhauser) stage but the scanning
range of the stage is not large enough to be able to access the entire plate. I am not sure
whether that is a physical limit or a software limit. It is worth checking this before you
purchase a plate adapter. (We did not, but we almost never use the plate adapter anyway.)

- As far as I know, plate adapters are not compatible with Leica's on-stage Z-scan unit.

I have a Leica SP5X with a Marzhauser motorized stage and a piezo Z-stage.
We bought a separate multi-well plate holder from Leica after the initial
purchase.
This works fine with the piezo z-stage. I haven't directly compared to confirm,
but I am fairly certain the z-range is similar to other stage inserts (just
depends on your sample).
If your stage travel range does not allow you to visit the whole area of the
multi-well plate, this can be adjusted in the software control of the microscope
and stage. Ask your Leica rep or contact me off-line.
I would also be happy to let you know the part number that I ordered from Leica.

Hello everyone,

We have a Leica SP5 X confocal microscope. We are planning to image  
96-well plates on it. But so far I haven't found anything to fit the  
plate on the stage.

I'm wondering anyone on the list has experiences with this kind of  
experiment. Any suggestion is welcome.

I'm really sure you will find the two insert system for your mmultiwell
plate.Directly from Leica. I bought them are:


Insert SuperZ     No : 11640416
For the Marzhauser plate  No : 11531434

Hi Yue,

There is the TISSUEscope slide scanner - that has confocal fluorescence imaging.  It has a very large stage because it can image large specimens/well plates.  It can hold a glass slide as large as 6x8".  www.huron-technologies.com
