You would probably want an AM ester derivative.  Beyond this, there are variables to consider:  What wavelength is preferred?  What wavelength options are on your confocal?  What Kd is desired?  Do you want ratiometric or non-ratiometric response?  Are other dyes present?

I obtained nice results with snarf 5f (pKa 7.2) in isolated skeletal muscle cells. This synthetic dye is ratiometric.
