slim stage top incubator

Hi all,

  I'm looking for a stage environmental chamber (temp and CO2) to keep cells alive while imaging on an inverted olympus. I am using oil immersion for the epi detection. In the forwards direction I am acquiring another signal via the use of a long working distance objective. It needs to have an NA around .6-.8 and the highest mag possible. It all works great on fixed cells but I fear that adding a chamber will increase the distance from the forward detection objective to a point where it will no longer work. What is the shortest  (along the optical axis) environmental chamber I can buy? As it stands even with a long working distance objective it is very close to the sample..

You do not have to sacrifice N.A. to image live cells.

Take a look at the variety of high numeric aperture compatible micro-
environmental systems from Bioptechs.

Temperature control, high N.A. compatibility, CO2, perfusion and more  
for your live-cell time-lapse imaging.

It helps us a lot if you fill out the user profile questionnaire and  
send it in so that we can properly identify the most appropriate  
system and accessories.

Jeff,

Are you collecting an image from above the sample?

If so I think simplest may be to enclose the microscope in an acyclic box
for temperature regulation.
eg. Solent, OKO Labs, PeCon.

Make your own sub-incubator to supply humidity, and feed this with a
premixed gas for the correct CO2. I use to use 5% CO2 mixed with air. You
could have a hole cut in the lid to accommodate your lens, and seal this
with a flexible sheet of rubber or silicone.

If I've got the idea of your setup completely wrong I apologise.

We can design an incubation system that will fit you needs. Even one
that would work with a dipping lens, if you would like to increase

Hi Jeff,

in our facility, we are pretty happy with the stage-top incubator system called
"Chamlide TC" by Live Cell Instruments, distributed by Quorum Technologies.
[No commercial interest, just a happy customer.] The chamber can
accommodate a number of different sample holder types (small glass-bottom
dishes, round cover glasses in a magnetic reusable holder, rectangular multi-
well dishes of slide size), and controls temperature, CO2 and humidity.
Since the imaging is through a regular cover glass (or glass-bottom dish), the
objectives on an inverted scope should work just like with a slide plus cover
glass. I'm not sure what you mean using the long working distance objective in
the "forward detection". Condensers can get pretty close to the top of the
incubator chamber, if that is your concern for the transmitted light setup. But
on our spinning disk systems (Quorum WaveFX on Leica DMI6000B), this
hasn't been a problem.
