My issue is that my sample cannot be mounted between a slide glass and cover slip. I should apply a treatment to the sample while I observe the sample. (My sample is wood cell wall.) I'm using glass bottom Petri Dishes from Mattek with good results, other compagnies are offering such petri dishes. (No commercial interest) cheers benoit

http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal Two options that come to mind are the MatTek type petri dishes http://www.glass-bottom-dishes.com/free-sample.html or have your machine shop make slide shaped blanks out of 1mm thick aluminum or plastic. Then you can cut or drill out any shape. We routinely cut a 15x15mm square notch at the middle of the slide opening onto one side of the slide. Now you can tape (or glue) a 22x22mm cover slip over the open area and use it on an inverted microscope with the cover slip below the sample and open above. You can tape another cover slip on the other side of the slide to have a coverslip-sample-coverslip sandwich with 1mm between coverslips. All this is predicated on the assumption that the slide does not need to be glass. I had good luck with using a 1/4" or 1/2" thick piece of plexiglass, with a 20mm hole drilled in the center, with coverslip glued over the hole using GE Silicone II sealant (most hardware stores carry that). The sealant cures in about a day, but remains flexible, so when the coverslip breaks, it is easy to scrape it off witjh a razor blade and glue a new one on.
