Dear All,

I would like to ask your advice about the situation we have with our Leica camera (DFC900 GT) on our our life cell Ca-Imaging system.

We noticed that we see crystals in our images (see the images for 20x and 100x, made without sample and without oil for 100x, in the attached link https://cloudstorage.elearning.uni-oldenburg.de/s/7fMgzCK3XnTGWbo ).

What I can see:

1. Crystals size does not change with magnification change (size stays the same for 20x, 40x, 63x, 100x).
2. When we turn the camera, the crystals do not rotate.
3. We can see the crystals even without objective, i.e. we remove an objective and have just ambient light shining, we still can see the crystals in the software.
4. We do not see crystals in the ocular.
5. When we opened the camera and looked at it, we could not see anything special, no matt film etc.

We asked Leica, they say that it is some crystallisation on the camera chip. The repair is about 11000€, new camera is about 16000€.

I would like to ask if you experienced a similar problem, how did you solve it?
Any ideas and suggestions are welcome!

Thank you in advance.

best wishes,
Vita

I know this is not quite the same but we had an issue with our Gatan camera
which is also a CMOS so it has a similar chip and the tech had us do an
annealing cycle to clean the chip, and then a new set of reference images
worked wonders to remove what noise was left. It might be worth a shot.

Good luck

Emma

Hi Vita,

We had a similar problem on a camera.  It turned out to be condensation which turned to ice.  We didn't get the camera repaired, but I turned off the camera cooling function and monitored the temperature. I only used it for brief periods, so this was a solution for me.

Good luck!!

Dear Vita,

the images look like condensation. Is the camera a cooled camera? Maybe the camera head has caught to much humdity, because if so, I mean if the scavengers are filled and/or the gas sealing was aging it might be that when switched on and the camera starts cooling the image sensor, then the humidity condensates and the images look similar.
I guess the camera needs a service to refresh the dry gas or the vacuum and dry it.

Best regards,
Gerhard

It is condensation.  https://www.flickr.com/photos/mcammer/35345268455/  We have had past discussions of this here.  Some cameras from the 1990s had to be attached to a vacuum pump periodically and resealed (I did this for someone a few years ago who was still using a Princeton Instruments camera with Windows 2000!)  But now cameras should not have the seal break.
It needs to be fixed, you need to move the scope to a dry room, or you need a new camera.

Hi Vita,
The "crystals" are very sharply focused, so it is not on the camera sensor itself.  It is somewhere further down the optical path.
Ice and frost usually have more definitely fractal edges in my experience.  Perhaps this is forming around fine particles somewhere in the optical setup.
A dessicant to dry it out and heating to room temperature or a bit above might help.
Check if your camera has a rechargeable desisccant cartridge or replaceable desiccant, in case it is forming on an optical window several mm in front of the sensor.
Cleaning may be needed as well.
Good luck,
Colin

Thank you in advance.

best wishes,
Vita

That’s what it looks like when the condensate is on the sensor.

For what it's worth, we had this issue with a 6 year old Hamamatsu Orca
Flash4.0 LT (albeit with a smaller area of condensation in the center of
the chip) and they replaced it for $3K. Seems reasonable to me; equivalent
to $500/yr extended warranty I’ve been quoted on similar cameras from other
vendors.

Dear Vita,

We had a similar problem with our CMOS on our High content system.
I concur with a few other replies that it is likely to be condensation.

In our case, the camera seal had eroded and it let moisture into the sensor array area.
The engineer said that resealing etc was not an option and a new camera was needed($$$)
We disabled the environmental control and left the camera on 24/7 to let the fan drive out the moisture.
Fortunately, this approach has worked so far for us.

Cheers

Yours sincerely,

Dear All,

I would like to ask your advice about the situation we have with our Leica camera (DFC900 GT) on our our life cell Ca-Imaging system.

We noticed that we see crystals in our images (see the images for 20x and 100x, made without sample and without oil for 100x, in the attached link https://cloudstorage.elearning.uni-oldenburg.de/s/7fMgzCK3XnTGWbo<https://cloudstorage.elearning.uni-oldenburg.de/s/7fMgzCK3XnTGWbo> ).

Thank you in advance.
